    Kyprolis vs Revlimid or Velcade

    I noted in my previous post that the preliminary results reported at Kyoto for kyprolis/melphalan/prednisone combo in NDMM patients under 65 years old: ORR was 89%, with 51% achieving greater than VGPR, compared to an ORR of 74%, with 33% achieving VGPR in the melphalan/prednisone/Velcade combo.
    What is even more interesting is that the results reported Palumbo et al in 2010 for the lenalidome/mephalan/prednisone combo in NDMM patients older than 65 years were ORR 75% and 32% VGPR, almost identical to the Velcade combo but clearly inferior to the kyprolis/melphalan/prednisone combo. Thus, although their underlying mechanisms are different, it would appear that Kyprolis is likely to show superior efficacy relative to Revlimid (and not just Velcade) in NDMM patients. However, it seems that Kyprolis has powerful synergistic effects when combined with Revlimid.
